ارزیابی تنوع ژنوتیپ‌های جو (.Hordeum vulgare L) بر اساس محتوای بتاگلوکان و نشانگرهای ISSR

در این مطالعه، تنوع محتوای بتاگلوکان دانه، در 20 لاین و رقم جو بر اساس طرح بلوک‌های کامل تصادفی در سه تکرار مورد بررسی قرار گرفت. هم­چنین، تنوع ژنتیکی بین این ژنوتیپ ­ها، با استفاده از نشانگر ISSR ارزیابی شد. استخراج بتاگلوکان به روش آنزیمی انجام گرفت. از نظر محتوای بتاگلوکان دانه در بین ژنوتیپ‌های جو، اختلاف معنی‌دار در سطح احتمال یک درصد وجود داشت. محتوای بتاگلوکان دانه بین 7.21 تا 12.48 درصد متغیر بود و بیشترین مقدار محتوای بتاگلوکان، در ژنوتیپ‌های یوسف، E94B3 و E94B17 اندازه‌گیری شد. آغازگرهای ISSR با متوسط درصد چندشکلی 69.79 درصد، تنوع ژنتیکی 0.25 و شاخص شانون 0.37 در بررسی تنوع ژنتیکی بسیار کارآمد عمل کردند. لاین‌ها و رقم‌های جو، در دو گروه مجزا با مطابقت زیاد با شجره ژنتیکی، از هم متمایز شدند. تعداد نه آغازگر دارای اطلاعات برای محتوای بتاگلوکان دانه توسط روش‌های ناپارامتری کروسکال-والیس، همبستگی اسپیرمن و تجزیه رگرسیون گام به گام شناسایی شدند. درصد تبیین تغییرات محتوای بتاگلوکان دانه توسط این آغازگرها، بین 24.3 تا 42.4 متغیر بود. آغازگرهای ISSR6(700)، ترکیب ISSR1+ISSR4(1400) و ترکیب IS2+ISSR2(1400) دارای ارتباط قوی‌تری با محتوای بتاگلوکان بودند. آغازگرهای دارای اطلاعات، امکان گزینش کارآمد ژنوتیپ‌های جو با محتوای بتاگلوکان بالاتر را فراهم می‌آورند.
