بررسی تنوع ژنتیکی گونه‎ هایی از جنس L. Allium بر اساس نشانگر ISSR در استان کردستان

جنس L. Allium، بهویژه زیر جنس Melanocrommyum شامل بخشهایی است که از لحاظ تاکسونومیکی بسیار پیچیده هستند. موقعیت سیستماتیکی گونههای هرکدام از بخشها، در طول زمان به‌دفعات بازبینیشده است. در مطالعهی حاضر تنوع ژنتیکی بین 32 اکوتیپ متعلق به 10 گونه مختلف از جنس Allium و ارتباط آن‌ها با استفاده از نشانگرهای ISSR مورد بررسی قرار گرفت. نه آغازگر مورد استفاده، 166 نوار چندشکل را با میانگین 18 نوار به ازای هر آغازگر تولید کردند. از بین آغازگرهای مورد استفاده، آغازگر ISSR873، با 27 نوار، بیشترین و آغازگر ISSR4 با 2 نوار کمترین تعداد نوار چندشکل را ایجاد نمودند. شاخص اطلاعات چندشکلی (PIC) نشانگرها بین 0.04 تا 0.43 متغیر بود. تجزیه خوشهای به روش UPGMA و تجزیه به مختصات اصلی، اکوتیپ‌های موردمطالعه را در چهار گروه قرار داد. نتیجه تجزیه خوشهای و تجزیه به مختصات اصلی نشان داد که بیشتر گونههایی که از لحاظ مورفولوژیکی به هم شبیه هستند در گروههای نزدیک به هم قرار گرفتند. بر اساس ضریب تشابه دایس، بیشترین درصد تشابه در بین اکوتیپهای Allium stipitatum و Allium saralicum (72 درصد) از زیر جنس Melanocrommyum و کمترین شباهت در بین اکوتیپهای گونههای Allium tripedale و Allium iranicum (12 درصد) بهدست آمد. اکوتیپهای با کمترین درصد تشابه به زیرجنسهای Allium و Nectaroscordum تعلق دارند که در خوشههای مجزا قرار گرفتند. بر اساس نتایج، اکوتیپهای بخشهای Pseudoprason، Melanocrommyum و Procerallium بیشترین قرابت را نشان دادند. بهطور کلی میتوان نتیجه گرفت که نشانگرهای ISSR برای طبقهبندی گونههای جنس Allium مفید بوده و پتانسیل کافی برای بررسیهای فیلوژنتیک گونهها را دارا هستند. به‌علاوه با توجه به نتایج مبنی‌بر وجود تنوع ژنتیکی قابل‌توجه در بین اکوتیپهای مطالعه شده از گونههای وحشی جنس Allium، از این تنوع میتوان در آینده در فرآیندهای بهنژادی گونههای زراعی بهره جست.
