پایداری هموپلاسمی و توارث سیتوپلاسمی داربست پروتئینی دارپین G3 در تکثیر زایشی و رویشی گیاهان ترانسپلاستوم توتون

مهندسی پلاستید مزایای بی‌شماری را برای نسل بعدی فناوری تراریختی گیاهی به ارمغان می‌آورد که شامل تسهیل انباشت تراژن‌ها و تولید سطوح بیان بالای پروتئین‌های نوترکیب می‌باشد. پروتئین‌های تکرار آنکرینی طراحی شده (دارپین‌ها) پروتئین‌های داربست غیرایمونوگلوبولینی نسبتاً کوچکی هستند که با تمایل بالا به اهداف اختصاصی خود متصل می‌شوند. G3 یک نوع دارپین طراحی شده برای اتصال به پروتئین گیرنده تیروزین‌کیناز HER2 (گیرنده 2 فاکتور رشد اپیدرمی انسان) است. در مطالعه قبلی یک فرآیند زیستی برای تولید دارپین G3 به‌عنوان عامل تصویربرداری در سرطان‌های با بیان بالای HER2 در سیستم بیان کلروپلاستی توتون تولید شد. در این مطالعه، پایداری هموپلاسمی و بیان ژن دارپین G3 در نسل رویشی و زایشی گیاهان T1 ترانسپلاستوم توتون مطالعه شد. تأیید حضور ژن دارپین G3 در گیاهان ترانسپلاستوم با استفاده از واکنش PCR انجام شد. آنالیز لکه‌گذاری سادرن وضعیت هموپلاسمی گیاهان ترانسپلاستوم را تأیید کرد. آنالیز لکه‌گذاری وسترن تجمع دارپین G3 را در کلروپلاست نسل بعدی گیاهان ترانسپلاستوم تأیید کرد. محتوی دارپین G3 تولید شده در کلروپلاست گیاهان ترانسپلاستوم با استفاده از الایزا برای پروتئین محلول کل کلروپلاست حدود 33 درصد برآورد شد. نتایج این مطالعه تأیید کرد که ژن دارپین G3 در گیاهان رویشی و زایشی نسل T1 گیاهان توتون ترانسپلاستوم به‌صورت پایدار و در سطوح بالا بیان می‌شود.
