شناسایی نشانگرهای میکروساتلیت پیوسته با نواحی ژنومی دخیل در مقاومت به جدایه های عامل بیماری پوسیدگی اسکلروتینیایی یقه ساقه در آفتابگردان روغنی (Helianthus annuus L.) در شرایط کنترل شده

آفتابگردان ‌(.Helianthus annuus L) ‌یک ‌محصول ‌مهم ‌زراعی ‌می‌‌باشد ‌که ‌روغن ‌آن ‌ارزش ‌غذایی ‌و ‌اقتصادی ‌بالایی ‌دارد. ‌پوسیدگی ‌پایه‌‌ی ‌ساقه ‌که ‌توسط ‌قارچ‌‌های ‌Sclerotinia sclerotiorum ‌و ‌S. minor ‌ایجاد ‌می‌‌شود، ‌یکی ‌از ‌بیماری‌‌های ‌مهم ‌و ‌مخرّب ‌آفتابگردان ‌است. ‌استفاده ‌از ‌ارقام ‌مقاوم ‌به ‌عنوان ‌مهمترین ‌و ‌موثرترین ‌روش ‌برای ‌کنترل ‌بیماری ‌در ‌نظر ‌گرفته ‌می‌‌شود. ‌در ‌پژوهش ‌حاضر، ‌ارتباط ‌و ‌پیوستگی ‌بین ‌30 ‌جفت ‌نشانگر ‌ریزماهواره ‌با ‌مقاومت ‌به ‌سه ‌جدایه ‌S. sclerotiorumو ‌سه ‌جدایه ‌S. minorدر ‌100 ‌لاین ‌آفتابگردان ‌با ‌استفاده ‌از ‌نرم‌‌افزارهای ‌Structure ‌و ‌Tassel ‌مورد ‌ارزیابی ‌قرار ‌گرفت. ‌بر ‌اساس ‌30 ‌جفت ‌نشانگر ‌ریزماهواره ‌مورد ‌استفاده ‌در ‌این ‌مطالعه، ‌ساختار ‌ژنتیکی ‌جمعیت ‌به ‌دو ‌زیر ‌جمعیت ‌(2=K) ‌تقسیم ‌گردید ‌که ‌نتایج ‌حاصل ‌از ‌رسم ‌بارپلات ‌نیز ‌مؤید ‌آن ‌بود. ‌در ‌تجزیه ‌ارتباط ‌با ‌استفاده ‌از ‌نرم‌‌افزار TASSEL ‌از ‌طریق ‌دو ‌روش GLM ‌و ‌MLM ‌، ‌به ‌ترتیب ‌14 ‌و ‌12 ‌نشانگر ‌SSRشناسایی ‌شد ‌که ‌ارتباط ‌معنی‌‌داری ‌با ‌نواحی ‌ژنومی ‌کنترل ‌کننده ‌مقاومت ‌به ‌بیماری ‌داشتند ‌و ‌تغییرات ‌قابل ‌توجهی ‌از ‌صفت ‌را ‌توجیه ‌نمودند. ‌شناسایی ‌نشانگرهای ‌مشترک ‌مانند ‌نشانگر ‌ORS617برای ‌جدایه‌‌های ‌M1 ‌از ‌گونه ‌S.minorو ‌J1 ‌از ‌گونهS.sclerotiorum ‌می‌‌تواند ‌ناشی ‌از ‌اثرات ‌پلیوتروپی ‌یا ‌پیوستگی ‌نواحی ‌ژنومی ‌دخیل ‌در ‌کنترل ‌مقاومت ‌به ‌این ‌جدایه‌‌ها ‌باشد. ‌به ‌دلیل ‌اینکه ‌این ‌نشانگرها ‌گزینش ‌همزمان ‌مقاومت ‌به ‌چندین ‌جدایه ‌قارچی ‌را ‌امکان‌‌پذیر ‌می‌‌سازد، ‌در ‌برنامه‌‌های ‌به‌‌نژادی ‌آفتابگردان ‌جهت ‌انتخاب ‌به ‌کمک ‌نشانگر ‌(MAS) ‌و ‌تولید ‌ارقام ‌مقاوم ‌به ‌بیماری ‌اهمیت ‌اساسی ‌دارد.
