تنوع آنالوگ‌های ژن‌های مقاومت به بیماری در ارقام مقاوم و حساس به زنگ گندم نان

بیماریهای قارچی به خصوص زنگهای زرد و قهوهای باعث کاهش عملکرد گندم در ایران و جهان میشوند. در این مطالعه، تنوع ژنتیکی 20 رقم گندم نان با پاسخ متفاوت به زنگهای قهوهای و زرد با استفاده از آغازگرهای طراحی شده براساس نواحی حفاظت شده ژنهای مقاومت به بیماری بررسی شد. الگوی نواری نشانگرهای چند شکل به صورت غالب امتیازدهی و تعداد نوار تکثیر شده و درصد چند شکلی تعیین گردید. علاوه بر این، برای هر ترکیب آغازگری، پارامترهای محتوای اطلاعات چند شکلی (PIC) و شاخص نشانگر (MI) محاسبه شد. از مجموع 11 ﺁغازگر منفرد و جفت، پنج ترکیب ﺁغازگری به همراه یک آغازگر منفرد تکثیر مناسب و قابل امتیازدهی تولید کردند. حداکثر و حداقل میانگین PIC به ترتیب مربوط به آغازگر LLOOP-1 و جفت آغازگر H2016-H2020 با میزان 0.50 و 0.28 بود. جفت آغازگرهای H2016-H1146 و H2016-H2020 با میانگین 4.80 و 2.84 به ترتیب حداکثر و حداقل MI را داشتند. تجزیه خوشهای براساس الگوریتم Neighbor-Joining و ضریب فاصله تکاملی P-distance، ارقام مورد مطالعه را به چهار گروه منتسب کرد و این گروهبندی با پاسخ به زنگ زرد ارقام مطابقت داشت. در تجزیه به بردارهای اصلی، پراکنش ارقام براساس دو بردار اصلی اول، چهار گروه حاصل از تجزیه خوشهای را تأیید کرد.
