مطالعه ارتباطی در سطح ژنوم صفات گیاهچه‌ای در ارقام گندم نان تحت شرایط نرمال و تنش شوری

به‌منظور شناسایی مکان ­های ژنی کنترل‌کننده صفات مورفو-فیزیولوژیک گیاهچه در 88 رقم گندم نان، آزمایشی در شرایط نرمال و تنش شوری 120 میلی ­مولار (معادل 12 دسی ­زیمنس بر متر)، در قالب طرح آلفا لاتیس ساده در گلخانه دانشکده کشاورزی دانشگاه ارومیه در سال زراعی 1400-1399 انجام شد. صفات میزان کلروفیل a، b و کل، کا روتنوئید، پرولین، وزن ‌تر و خشک گیاهچه، طول گیاهچه، محتوای نسبی آب برگ (RWC) و غلظت یون‌ها (سدیم، پتاسیم و نسبت پتاسیم به سدیم) اندازه‌گیری شدند. پس از ژنوتیپ‌سنجی به‌وسیله توالی‌یابی با فنّاوری Ion Torrent و حذف SNPهایی با بیش از 20 درصد داده گمشده و فراوانی آلل جزئی کمتر از 5 درصد، تعداد 5869 SNP شناسایی شد. بر اساس نقشه ­یابی ارتباطی با روش مدل خطی مخلوط (MLM) برای شرایط نرمال در مجموع 25 ارتباط نشانگر-صفت (MTA) شناسایی شد. ژنوم A بیشترین و ژنوم D کمترین تعداد MTA را به خود اختصاص دادند. در بین صفات مورد بررسی در شرایط نرمال، کلروفیل a بیشترین تعداد MTA را روی کروموزوم­ های 1A، 3B، 3D، 5B و 7A نشان داد و برای شرایط تنش شوری 21 MTA شناسایی شد که ژنوم B بیشترین تعداد MTA و ژنوم D کمترین تعداد MTA را به خود اختصاص دادند. برای وزن ‌تر گیاهچه پنج MTA شناسایی شد که روی کروموزوم ­های 4A و 6B قرار داشتند. نتایج پژوهش حاضر، اطلاعات ارزشمندی در ارتباط با مکان­ های ژنی مرتبط با صفات مورد مطالعه ارائه می­دهد که می­ توان پس از تأیید در جمعیت­ های دو والدی و آزمایش‌های تکمیلی، در برنامه­ های به­نژادی گندم در گزینش به کمک نشانگر از این یافته‌ها استفاده نمود.
