ارزیابی برخی صفات بیوشیمیایی و بیان دو ژن از خانواده MYB در دو رقم گندم تحت تنش خشکی

تنش‌های محیطی از عوامل اصلی کاهش دهنده رشد و نمو و عملکرد گیاهان زراعی به‌شمار می­آیند و همواره امنیت غذایی انسان‌ها را تهد‌ید می‌کنند. این مطالعه به­منظور بررسی اثر تنش خشکی بر تغییرات صفات بیوشیمیایی و میزان بیان ژن‌های عوامل رونویسی MYB در دو رقم گندم (تجن و زاگرس)، تحت تنش خشکی انجام شد. آزمایش به‌صورت فاکتوریل در قالب طرح کاملاً تصادفی با 3 تکرار انجام شد. اعمال تیمار خشکی در سه سطح 40، 70 و 100 درصد ظرفیت زراعی و 4 هفته پس از جوانه‌زنی انجام شد. 20 روز پس از اعمال تنش نمونه‌برداری از برگ و ریشه به­منظور بررسی بیان ژن­ها و برخی صفات بیوشیمیایی صورت گرفت. نتایج بررسی محتوای کلروفیل تحت تنش نشان داد که با افزایش شدت تنش در ژنوتیپهای مختلف محتوای کلروفیلa  و b کاهش می‌یابد. میزان کاهش کلروفیلa  و b در ژنوتیپ تجن در تنش شدید، بیشتر از ژنوتیپ زاگرس بود. همچنین محتوای TBARM تحت تنش شدید خشکی به‌طور معنی‌داری بیشتر از وضعیت تنش متوسط بود و این افزایش در ژنوتیپ تجن بیشتر از رقم زاگرس دیده شد. تجزیه و تحلیلRT-PCR  نشان داد که ژنهای خانواده MYB مورد مطالعه، تحت تنش خشکی افزایش بیان نشان می‌دهند. از طرفی رقم زاگرس که جزو ارقام متحمل به تنش خشکی می‏باشد برای تمامی ژن‏های مورد بررسی افزایش بیان بیشتری نسبت به رقم تجن داشت که امکان استفاده از این رقم را در بحث برنامه­های بهنژادی فراهم می­کند. همچنین با توجه به اهمیت ژن­های خانواده  MYB طی تنش خشکی، از یافته‌های پژوهش حاضر می‌توان در برنامه‌های به­نژادی و هرمی کردن ژن­ها استفاده کرد.
