ارزیابی تنوع ژنتیکی و شناسایی ارتباط خصوصیات دانه با نشانگر‌های SSR و SCoT در لوبیا

تحقیق حاضر به‌منظور ارزیابی تنوع ژنتیکی 18 رقم و لاین امیدبخش لوبیا و شناسایی نشانگرهای آگاهی‌بخش SSR و SCoT مرتبط با 14 خصوصیت دانه شامل تعداد دانه در غلاف، وزن 100 دانه، طول دانه، عرض دانه، پروتئین خام، قند محلول کل، نشاسته، میزان چربی، خاکستر، غلظت آهن، کلسیم، منیزیوم، روی و اسید اورونیک انجام شد. میزان محتوای چندشکلی (PIC) برای نشانگر SSR از 0.2 تا 0.5 با متوسط 0.39 و برای نشانگر SCoT بین 0.19 تا 0.42 با میانگین 0.34 متغیر بود. میانگین کل قدرت تفکیک نشانگر SSR و SCoT به‌ترتیب برابر 1.54 و 5.34 بود که می‌تواند بیانگر مناسب‌تر بودن نشانگر SCoT نسبت به نشانگر SSR برای بررسی تنوع ژنتیکی در ژنوتیپهای مورد مطالعهی لوبیا باشد. با دندروگرام حاصل از تجزیه خوشه‌ای با استفاده از هر دو نشانگر، ژنوتیپهای لوبیا به سه گروه متفاوت دستهبندی شدند. تجزیه به مختصات اصلی برای نشانگر SSR نشان داد که مجموع تغییرات توجیه شده توسط دو مؤلفه اول برابر با 59.05 درصد و برای نشانگر SCoT تنها 25.43 درصد بود که این موضوع می‌تواند نشاندهنده پراکنش بهتر نشانگرهای SCoT نسبت به نشانگرهای SSR در سطح ژنوم لوبیا باشد. تجزیه واریانس مولکولی بر اساس گروهبندی حاصل از تجزیه خوشهای نشان داد که تنوع داخل گروه بر اساس نشانگرهای SSR و SCoT به‌ترتیب برابر با 89 و 78 درصد بود. نتایج تجزیه رگرسیون بین نشانگرهای مورد بررسی و خصوصیات دانه لوبیا نشاندهنده‌ی وجود ارتباط معنی‌دار هر نشانگر با چندین صفت مورد بررسی بود که این موضوع می‌تواند بیانگر ارتباط یا پیوستگی مکانهای نشانگری باشد. دوازده صفت از 14 خصوصیت مورد بررسی دانه لوبیا با حداقل یک نشانگر مولکولی ارتباط معنی‌دار نشان دادند.
