تحلیل ژنومی خانواده ژنی WOX در کنجد (.Sesamum indicum L) و بررسی الگوی بیان برخی از ژن‌های آن تحت تنش‌ خشکی

چکیده
در این پژوهش، شناسایی جامع و تحلیل خانواده ژنی WOX در گیاه کنجد (Sesamum indicum L.) انجام شد. در مجموع ۳۶ ژن SiWOX در ژنوم کنجد شناسایی گردیدند که به‌طور نامتقارن بر روی کروموزوم‌ها توزیع شده و بیشترین تراکم در گروه‌های پیوستگی (کروموزوم‌های) 1 و 2‌ مشاهده شد. بررسی ویژگی‌های فیزیکوشیمیایی نشان داد که طول پروتئین‌های WOX بین 175 تا 853 اسیدآمینه، وزن مولکولی بین 11/20 تا 62/93 کیلو دالتون و شاخص آلیفاتیک بین 50 تا 55/92 متغیر بود. مقادیر pI بین 99/4 تا 56/9 بوده و شاخص منفی GRAVY ماهیت آب‌دوست پروتئین‌ها را نشان داد. پیش‌بینی جایگاه سلولی بیانگر آن بود که اغلب پروتئین‌ها در هسته قرار دارند، اما برخی نیز در کلروپلاست، سیتوپلاسم، میتوکندری و غشای پلاسمایی مشاهده شدند. تحلیل فیلوژنتیکی ژن‌های SiWOX آن‌ها را در چهار گروه اصلی طبقه‌بندی کرد. بررسی عناصر سیس در ناحیه پروموتری، ۶۱ عنصر تنظیمی مرتبط با پاسخ‌های نوری، هورمونی و تنش‌های محیطی را آشکار نمود. نتایج تحلیل ساختار ژنی، تفاوت در تعداد اگزون‌ها و اینترون‌ها را نشان داد و وجود دمین حفاظت‌شده هومودمین در تمام پروتئین‌ها نقش آن‌ها در اتصال به DNA را تأیید کرد. تحلیل بیان ژن‌های SiWOX9، SiWOX16 و SiWOX36 در دو ژنوتیپ سرداری و دشتستان نشان داد که ژنوتیپ سرداری پاسخ بیشتری به تنش دارد؛ به‌ویژه برای ژن SiWOX16 که افزایش چشمگیری در 48 ساعت نشان داد. در مقابل، در ژنوتیپ دشتستان بیان این ژن‌ها کاهش یا نوسان خفیف داشت. با این حال، برای شناسایی ژن‌های کاندید برای بهنژادی ارقام متحمل به تنش و فراهم کردن پایه‌ای برای مطالعات عملکردی آینده و بهنژادی به کمک ابزارهای ژنومی، تحقیقات بیشتر به‌ویژه بر اساس داده‌های توالی‌یابی RNA ضروری است.
