بررسی تغییرات بیان مهم‌ترین ژن‌های درگیر در تجمع ساکارز در لاین‌های نیشکر تراریخت دارای ژن موتان‌سوکروز

چکیده
نیشکر (Saccharum officinarum) یکی از گیاهان صنعتی مهم دنیا است که مقادیر زیادی ساکارز را در ساقه‌ها ذخیره میکند. ساکارز سوبسترای اصلی آنزیم‌های کلیدی مسئول سنتزِ شمار زیادی از بیوپلیمرها ارزشمند صنعتی است. آنزیم موتان‌سوکروز (GtfI) با استفاده از ساکارز، بیوپلیمر صنعتی-پزشکی موتان (Mutan) را سنتز میکند. در نتیجه، انتظار می‌رود که بیان ژن gtfI، علاوه‌بر تبدیل بخشی از ساکارز ساقه به موتان، سبب تغییر الگوهای بیان ژن‌های درگیر در بیوسنتز ساکارز سلولهای نیشکر شود. در این پژوهش، سطوح بیان ژن‌های سوکروز فسفات سنتاز (Sps)، سوکروز سنتاز (Susy)، سولوبل اسید اینورتاز (Sai) و اینورتاز (Inv) به‌عنوان ژن‌های اصلی کنترل‌کننده مسیرهای سنتز و تجمع ساکارز، در هر دو بافت برگ و قلمه ساقه در سه لاین تراریخته (B9، B11 و B38) بیان‌کننده ژن موتان‌سوکروز باکتریایی و شاهد غیرتراریخت با استفاده از روش Semi-quantitative RT-PCR مورد ارزیابی قرار گرفت. نتایج این مطالعه نشان داد که بیان ژن‌های درگیر در سنتز و تجمع ساکارز در لاینهای تراریخت، تحت تأثیر افزایش بیان ژن موتان‌سوکروز قرار گرفتند. به‌طور مشخص، سطوح رونویسی ژن Sps در گیاهان تراریخت نسبت به گیاه شاهد افزایش یافت. در مقابل، بیان ژن Susy تفاوت چندانی را بین لاین‌های تراریخت و شاهد نشان نداد. برعکس، میزان رونویسی ژن‌های Sai و Inv در گیاهان شاهد بیشتر از لاین‌های تراریخت بود. در میان لاینهای تراریخت مورد ارزیابی، لاین B38 بیشترین تغییرات را در بیان ژنها از خود نشان داد؛ بنابراین، دست‌ورزی‌های ژنتیکی هدفمند با هدف بیش‌بیان (Overexpression) ژن‌های درگیر در بیوسنتز ساکارز، به‌طور بالقوه می‌تواند در دسترس بودن سوبسترای ساکارز را افزایش داده و از این طریق، آنزیم موتان‌سوکروز را جهت تولید بهینه‌ی بیوپلیمر موتان تغذیه کند.
