بررسی‌ تنوع‌ ژنتیکی‌ برخی‌ از گونه‌های‌ جنس Salvia منطقه زاگرس مرکزی با استفاده‌ از نشانگر مولکولی‌ iPBS

چکیده
جنس Salvia از مهم‌ترین جنس­های گیاهان دارویی تیره نعناعیان است که به‌دلیل دارا بودن ترکیبات زیست‌فعال و ارزش دارویی، نیازمند شناسایی و حفاظت ذخایر ژنتیکی می‌باشد. استفاده از نشانگرهای مولکولی مبتنی بر رتروترانسپوزون‌ها، به‌ویژه نشانگر iPBS، به‌دلیل توانایی بالا در آشکارسازی چندشکلی ژنتیکی، رویکردی کارآمد برای ارزیابی تنوع ژنتیکی گیاهان دارویی محسوب می‌شود. در این پژوهش تنوع ژنتیکی و ساختار جمعیتی ۵۰ اکوتیپ از هفت گونه جنس Salvia، منطقه زاگرس مرکزی ایران با استفاده از ۱۰ آغازگر iPBS بررسی شد. نتایج نشان داد که نشانگرهای iPBS توانایی بالایی در آشکارسازی تنوع ژنتیکی گونه‌های مورد مطالعه داشتند؛ به‌طوری‌که میانگین درصد نوارهای چندشکل 33/98 درصد و میانگین شاخص اطلاعات چندشکلی (PIC) برابر 36/0 به دست آمد. تحلیل خوشه‌ای بر اساس ضریب تشابه دایس و روش Neighbor-Joining، ۵۰ ژنوتیپ مورد مطالعه را در گروه‌های ژنتیکی متمایز تفکیک کرد و آزمون مانتل نیز با مقادیر 68/0r = و مقدار احتمال بسیار معنی‌دار (002/۰p =) همبستگی بین ماتریس تشابه ژنتیکی و الگوی خوشه‌بندی را نشان داد. نتایج تحلیل تنوع جمعیتی نشان داد که جمعیت‌های S. macrosiphon و S. spinosa دارای بالاترین سطح تنوع ژنتیکی در بین جمعیت‌های مورد مطالعه بودند. وجود این سطح بالای تنوع ژنتیکی، این جمعیت‌ها را به‌عنوان منابع ژرم‌پلاسم ارزشمند برای برنامه‌های بهنژادی معرفی می‌کند. در مقابل جمعیت‌های S. reuterana با کمترین میزان تنوع شاخص‌های ژنتیکی نسبت به سایر جمعیت‌ها، یک ذخیره ژنتیکی متمایز محسوب می‌شود که نیازمند توجه ویژه در برنامه‌های حفاظت و نگهداری ژرم‌پلاسم است. همچنین، گونه­های S. multicaulis و S. limbata به‌دلیل قرارگیری در شاخه‌های مستقل دندروگرام و تمایز ژنتیکی بالا، می‌توانند نماینده گروه‌های ژنتیکی منحصر‌به‌فردی باشند که حفاظت آن‌ها برای حفظ تنوع ژنتیکی جنس Salvia اهمیت دارد.
