چکیده
دیفنسینها (Defensins) یکی از مهمترین خانوادههای پپتیدهای ضدمیکروبی گیاهی هستند که نقش کلیدی در دفاع گیاهان در برابر عوامل بیماریزا ایفا میکنند. این پژوهش با هدف شناسایی، توالییابی و بررسی ویژگیهای ساختاری، تکاملی و عملکردی اعضای خانواده ژنی دیفنسین در سه رقم لوبیای معمولی (Phaseolusvulgaris L.) انجام گرفت. ژنهای دیفنسین با استفاده از جستجوی همولوژی در ژنوم مرجع لوبیا شناسایی و آغازگرهای اختصاصی طراحی شدند. پس از استخراجDNA، ژنها با PCRتکثیر و محصولات به روش سنگر تعیین توالی شدند. ویژگیهای ژنی و پروتئینی با ابزارهای مختلف بیوانفورماتیکی بررسی و روابط فیلوژنتیکی، خاصیت ضدمیکروبی احتمالی و الگوی بیان ژنها در شرایط مختلف تحلیل شد. نتایج این مطالعه نشان داد که تعداد ده ژن دیفنسین در ژنوم لوبیا وجود دارد که بر روی چهار کروموزوم مختلف قرار گرفتهاند. توالی ناحیه کدکننده این ژنها در هر سه رقم کاملاً یکسان بود. تمامی پروتئینها دارای دومین اختصاصی Knot1، سیگنالپپتید ترشحی، هشت سیستئین حفاظتشده و موتیف γ-coreبودند. نتایج تجزیه وتحلیل صورت گرفته همچنین ظرفیت بالای فعالیت ضدمیکروبی این پروتئینها را تأیید کرد. تحلیل فیلوژنتیکی و بررسی بیان ژنها در شرایط in silicoنشان داد که اعضای این خانواده دارای الگوهای تکاملی و تنظیمی متفاوت بوده و ژنهای PvDef3و PvDef8بیشترین پاسخ را به تنشهای زیستی و غیرزیستی نشان دادند. نتایج این پژوهش اطلاعاتی مفید در مورد خانواده ژنی دیفنسین در لوبیا فراهم کرده و میتواند در بهنژادی ارقام مقاوم به بیماریها و توسعه کاربردهای زیستفناورانه مورد استفاده قرار گیرد.
